IMPORTANT DISCLAIMER: This article is for educational and informational purposes only. Phenibut is sold strictly as a research chemical for laboratory and scientific research use only. It is NOT intended for human consumption, animal consumption, or any clinical applications. This product is designated for use by qualified researchers in controlled laboratory settings.

Phenibut is a synthetic analog of gamma-aminobutyric acid (GABA), the main inhibitory neurotransmitter of the central nervous system. Its chemical name is 4-amino-3-phenylbutanoic acid, also written β-phenyl-GABA: a GABA backbone with a phenyl ring on the β-carbon. In published binding studies it acts at 2 targets, the GABA-B receptor and the α2-δ subunit of voltage-dependent calcium channels. Research suppliers, including Nordic Chems, sell it as the hydrochloride salt, Phenibut HCL (CAS 3060-41-1).

This overview covers the identity data for both forms of the compound, how it differs from GABA, the receptor constants reported in the literature, how laboratories use it, the quality documents to check, and its legal status.

What Is Phenibut? The Chemical Foundation

Phenibut was first synthesized in the Soviet Union by V. V. Perekalin and colleagues at the Herzen Pedagogic Institute in Leningrad, now St. Petersburg. The World Health Organization pre-review report on phenibut cites a 1954 publication for the synthesis, and a 2001 review of the published literature dates its pharmacological characterization to the 1960s. Early papers called it phenigamma. Fenibut and β-phenyl-GABA are other names used in the literature.

The compound is sold in 2 forms, the free amino acid and the hydrochloride salt. Their formulas, molecular weights and CAS numbers differ, so confirm which form a method, a certificate or a label refers to before calculating a concentration.

Property Phenibut (free amino acid) Phenibut HCL (hydrochloride)
Chemical name 4-amino-3-phenylbutanoic acid 4-amino-3-phenylbutanoic acid hydrochloride
Molecular formula C10H13NO2 C10H14ClNO2

The hydrochloride carries 1 hydrogen chloride unit per phenibut unit, so 215.67 g/mol is the figure to use when preparing a molar solution from Phenibut HCL. Our Phenibut HCL vs FAA comparison works through the mass, solubility and pH differences between the 2 forms.

How Phenibut Differs From GABA

GABA is a 4-carbon amino acid. Phenibut keeps that backbone and adds a phenyl ring at the β-carbon. The ring makes the molecule more lipophilic than GABA, and the literature attributes phenibut's passage across the blood-brain barrier in animal models to that change, where GABA itself crosses poorly. Baclofen, the reference compound in the binding studies below, is the same structure with a chlorine atom at the para position of the ring. Our guide to the chemical structure of phenibut covers the molecular architecture in more depth.

Stereochemistry: R-Phenibut And S-Phenibut

The β-carbon is a chiral center, so phenibut exists as 2 mirror-image forms. The WHO pre-review describes R-phenibut as the biologically active isomer and lists separate CAS numbers for the R and S hydrochlorides: 52992-48-0 and 52950-37-5. Material sold under 3060-41-1 or 1078-21-3 is the racemate, an equal mixture of both, unless its certificate states otherwise. Standard HPLC does not separate the 2 isomers. That takes a chiral method.

What Does Phenibut Bind To? Published Receptor Data

The 2001 review describes phenibut as a GABA-mimetic that acts primarily at GABA-B receptors and, to some extent, at GABA-A receptors. Later radioligand studies put numbers on 2 targets: GABA-B receptor binding measured against the antagonist radioligand [3H]CGP54626, and binding at the α2-δ subunit of voltage-dependent calcium channels, measured with radiolabelled gabapentin in rat brain membrane preparations.

Compound GABA-B receptor α2-δ subunit
Racemic phenibut 177 ± 2 µM -
R-phenibut 92 ± 3 µM 23 µM
S-phenibut did not bind 39 µM
Baclofen 6.0 ± 1 µM 156 µM
Gabapentin - 0.05 µM

Values are in vitro binding constants as reported by Dambrova et al. (2008) and Zvejniece et al. (2015). A lower number means tighter binding. A dash means the cited abstract gives no value.

The table supports 3 working conclusions:

  • Racemic phenibut binds the GABA-B receptor roughly 30 times more weakly than baclofen (177 µM against 6.0 µM).
  • R-phenibut binds the α2-δ subunit, the site gabapentin binds, about 4 times more tightly than it binds GABA-B (23 µM against 92 µM).
  • S-phenibut binds α2-δ and does not bind GABA-B, so the 2 isomers are not interchangeable in a receptor study.

For experimental design, an effect observed with racemic phenibut cannot be assigned to the GABA-B receptor without a control. A selective GABA-B antagonist, or a side-by-side run with baclofen and gabapentin, separates the 2 targets.

How Laboratories Use Phenibut

Documented laboratory uses fall into 4 groups:

  • Receptor binding assays: competition against a radioligand to measure affinity at GABA-B receptors or the α2-δ subunit, as in the studies above.
  • Structure-activity comparisons: phenibut, baclofen, gabapentin and GABA share a GABA backbone and differ in the group attached to it, which makes them a standard comparison set.
  • Enantiomer studies: R-phenibut and S-phenibut tested against the racemate to assign an effect to 1 isomer.
  • Analytical reference work: identity confirmation, method development, calibration and validation by HPLC or LC-MS.

Disposition data are thinner than the receptor data. The WHO pre-review notes that the commonly repeated statements about how phenibut is metabolized and cleared trace back to the 2001 review and could not be verified against primary references, so treat them as unconfirmed.

Experimental Design Considerations

  • Concentration: record concentrations in molar units, and write the form (HCL or free amino acid) and its CAS number beside every value. Published binding constants sit in the micromolar range.
  • Controls: include a vehicle control, a reference agonist such as baclofen, and a GABA-B antagonist where receptor assignment matters.
  • Time course: set exposure durations and measurement time points before the run.
  • Replication: plan enough independent replicates for the statistics the study needs.

Quality Documents To Check For Research-Grade Phenibut HCL

A certificate of analysis (COA) for phenibut should show:

  • A batch or lot number that matches the container
  • The test date
  • The method used, such as HPLC, mass spectrometry or LC-MS
  • The purity result
  • Identity confirmation and the form tested (HCL or free amino acid)

Nordic Chems requires every batch to clear 99% purity at an independent laboratory before it goes into production, with HPLC and mass spectrometry testing by BioRegen Labs in Houston, Texas and Janoshik Analytical in Prague, Czech Republic. Each certificate carries the batch number and the test date, and emailing support@nordicchems.is with an order number identifies the batch a shipment came from. The quality control page shows our third-party test results.

For work built around 1 isomer, ask for chiral HPLC data. Routine purity and identity tests confirm the phenibut molecule and do not report the ratio of R to S.

Storage And Handling

Phenibut HCL is hygroscopic, meaning it takes up moisture from the air. Keep it in a tightly sealed container in a cool, dry place away from direct light, and reseal the container promptly after each use. Handle the powder with a lab coat, gloves and eye protection, transfer it under local exhaust or in a fume hood, and dispose of waste through your institution's chemical waste stream. The safety data sheet (SDS) and the supplier's storage guidance take precedence over general advice. The Nordic Chems FAQs cover ordering and shipping questions.

Legal Status Of Phenibut As Of October 2026

The legal status of phenibut depends on jurisdiction, and Utah and Connecticut have both scheduled it since May 2025.

  • United States, federal: phenibut is not a federally controlled substance and has no approved use. In April 2019 the FDA stated that it does not meet the legal definition of a dietary ingredient and issued 3 warning letters to companies selling it in consumer products.
  • Alabama: Schedule II under Ala. Code 20-2-25.
  • Utah: Schedule I under H.B. 173, effective May 7, 2025.
  • Connecticut: Schedule I, effective March 25, 2026.
  • Outside the United States: Australia has classified phenibut as a Schedule 9 prohibited substance since 2018. The WHO pre-review, published in 2021, lists Hungary, Italy, Lithuania and France as controlling it, and notes that it holds a pharmaceutical registration in Russia. At the time of that review it was not listed under the United Nations control conventions.

Nordic Chems does not ship phenibut to Alabama, Connecticut or Utah. Rules change, so confirm the current position for your state or country before ordering. This section is general information and is not legal advice. Our phenibut regulatory compliance guide covers labeling, claims and documentation in more detail.

Summary

Phenibut is β-phenyl-GABA, a GABA analog with measured micromolar affinity for GABA-B receptors and for the α2-δ subunit of voltage-dependent calcium channels, with GABA-B binding carried by the R isomer. For laboratory work, 4 points matter most: know which form is in the container (Nordic Chems supplies the hydrochloride, CAS 3060-41-1, 215.67 g/mol), work in molar units, hold a batch-matched certificate, and check state rules before ordering.

Research use only. Phenibut HCL from Nordic Chems is supplied strictly for laboratory research and analytical purposes. It is not for human or animal consumption, ingestion, or any medical, dietary or therapeutic use, and it has not been evaluated or approved by the FDA. Nothing in this article is guidance for any use outside a controlled research setting.

FAQs

What is phenibut?

Phenibut is a synthetic analog of the neurotransmitter GABA with the chemical name 4-amino-3-phenylbutanoic acid. Structurally it is GABA with a phenyl ring on the β-carbon. In published binding studies it acts at GABA-B receptors and at the α2-δ subunit of voltage-dependent calcium channels. Nordic Chems supplies it as Phenibut HCL for laboratory research only.

What receptors does phenibut bind?

Radioligand studies report GABA-B affinity constants of 177 µM for racemic phenibut and 92 µM for R-phenibut, against 6.0 µM for baclofen. R-phenibut and S-phenibut bind the α2-δ subunit of voltage-dependent calcium channels at 23 µM and 39 µM. S-phenibut did not bind GABA-B.

What is the difference between phenibut HCL and phenibut FAA?

Phenibut HCL is the hydrochloride salt: C10H14ClNO2, 215.67 g/mol, CAS 3060-41-1. Phenibut FAA is the free amino acid: C10H13NO2, 179.22 g/mol, CAS 1078-21-3. The phenibut structure is the same in both, and the mass needed for a given molar concentration differs. Nordic Chems stocks the hydrochloride.

How should research-grade phenibut HCL be stored?

Store it in a tightly sealed container in a cool, dry place away from direct light. Phenibut HCL is hygroscopic, so limit the time the container is open. Follow the safety data sheet and your institution's chemical storage rules.

What analytical methods are used to characterize phenibut?

Common techniques are HPLC for purity, LC-MS or GC-MS for identity, and NMR and IR spectroscopy for structure. Separating R-phenibut from S-phenibut requires a chiral method such as chiral HPLC.

Is phenibut a controlled substance in the United States?

Phenibut is not a federally controlled substance as of October 2026. Alabama (Schedule II), Utah (Schedule I, effective May 7, 2025) and Connecticut (Schedule I, effective March 25, 2026) control it under state law, and Nordic Chems does not ship phenibut to those states. Check the current rules for your jurisdiction before ordering.
