Tianeptine is used in laboratory work as a mu-opioid receptor agonist, not as a serotonergic agent. That distinction decides whether a study design holds up. Research-grade tianeptine is supplied to qualified laboratories for preclinical depression models, stress neurobiology, and synaptic plasticity work, and it is not for human or animal consumption. Research-grade tianeptine reaches the bench in 3 salt presentations, and each one carries a different molar mass, which is where most concentration errors start. This guide covers the receptor evidence, the salt-form arithmetic, the analytical data a usable lot certificate has to show, and where the United States regulatory position stood in September 2026.

POWDER USED FOR RESEARCH PURPOSES

What is Research-Grade Tianeptine?

Research-grade tianeptine is a dibenzothiazepine chemical supplied at a verified purity specification, with lot-specific analytical data, strictly for laboratory and educational research. It is characterized by CAS number, PubChem identifier, molecular formula and molar mass on a per-lot certificate. It is not a food, not a dietary product, and not approved for any human application.

Nordic Chems supplies it in 3 presentations: sulfate capsules at 25 mg, 50 mg, 100 mg and 350 mg, sodium capsules at 12.5 mg, and sodium powder in a 10 gram format. A 50 mg/ml sodium solution is also listed. Identity data is published against PubChem's tianeptine record, and every lot carries its own certificate rather than a generic specification sheet.

Is Tianeptine a Serotonergic Compound?

No. Tianeptine was described for decades as a serotonin reuptake enhancer, and that model no longer holds. Gassaway and colleagues reported in Translational Psychiatry in 2014 that tianeptine acts at the mu-opioid receptor, with agonist activity in the sub-micromolar range and no meaningful affinity at the serotonin transporter. Samuels and colleagues followed in Neuropsychopharmacology in 2017, showing that the behavioral signal disappears in mu-opioid receptor knockout animals. A 2021 paper in the same journal narrowed the site further, to mu-opioid receptors on hippocampal GABAergic interneurons.

Here is the part that matters at the bench, and that almost no page on this topic says out loud. If a comparative study pairs tianeptine against a serotonergic reference compound on the assumption that both act on the same system, the control is not a control. It is a second experiment asking a different question. Study designs written before 2014, and the many secondary summaries that still repeat the reuptake-enhancement description, will produce results that cannot be read as a comparison. Check the receptor assumption in the protocol before ordering anything.

The serotonin literature is not worthless. It is historical. Read pre-2014 serotonergic framing as a record of what the field believed, and design against the receptor evidence that replaced it.

The Receptor Profile Behind the Research Signal

Tianeptine's usefulness in laboratory models comes from acting on 2 systems at once. Mu-opioid receptor agonism is the initiating event. Downstream, the compound alters glutamatergic signaling, with documented effects on AMPA and NMDA receptor function in animal preparations, and this is the arm that connects to synaptic plasticity endpoints. The combination is unusual, and it is why the compound keeps appearing in work on neurochemical pathways that other compounds do not reach the same way.

For study design, the sequence matters. Receptor-level effects appear first, glutamatergic and structural changes follow over longer exposure periods. A protocol measuring only a structural endpoint at a single late timepoint will miss the mechanism that produced it.

Which Preclinical Depression models use Tianeptine?

Tianeptine appears most often in 4 model types: behavioral despair paradigms such as the forced swim test, chronic stress protocols, social defeat paradigms, and hippocampal slice electrophysiology. Each one reads a different part of the same mechanism.

Behavioral despair paradigms measure immobility and swimming patterns in rodents. Chronic stress protocols run over weeks and pick up the adaptive changes that a single-exposure design cannot. Social defeat paradigms read the stress axis rather than the compound directly, which makes them useful for separating a receptor effect from a stress-response effect. Hippocampal slice work measures long-term potentiation and synaptic transmission directly, and given the 2021 interneuron finding, slice preparations are now the closest read on the actual site of action.

Stress-axis endpoints run alongside all 4: corticosterone concentration, adrenal weight, cytokine expression, microglial activation, and markers of oxidative stress such as antioxidant enzyme activity and lipid peroxidation.

Salt Form and Molar Mass, Where 5% Errors begin

Tianeptine free acid has a molar mass of 437.0 g/mol. The sodium salt is 458.9 g/mol. Calculating a concentration from the free acid figure while weighing out the sodium salt overstates the amount administered by about 5%, because the sodium salt is only 95.2% tianeptine by mass. On a 10 mg weighing, that is a 9.52 mg free acid equivalent, not 10 mg.

Form CAS PubChem CID Molecular formula Molar mass
Tianeptine sodium 30123-17-2 23663953 C21H24ClN2NaO4S 458.9 g/mol
Tianeptine hemisulfate monohydrate 1224690-84-9 45375836 C42H56Cl2N4O14S3 Take from the lot certificate

The sulfate row needs a warning. Published molar masses for CAS 1224690-84-9 do not agree across chemical reference databases, because sources differ on whether the compound is expressed as the 1:1 sulfate or the hemisulfate monohydrate. The two are not interchangeable in a calculation. Do not take a sulfate molar mass from a database. Take it from the certificate that came with the lot in hand, and record which salt the certificate names.

State the salt form in the methods section, every time. A paper that says "tianeptine, 10 mg" without naming the salt cannot be replicated, and reviewers increasingly send those back.

Neuroplasticity Endpoints Worth Measuring

The structural arm of tianeptine research concentrates on 3 endpoints: dendritic spine density and morphology in hippocampal tissue, BDNF expression across brain regions, and synaptic protein markers such as PSD-95 and synaptophysin. Receptor binding assays and signal transduction work on MAPK/ERK, PI3K/Akt and CREB phosphorylation sit underneath them.

Microdialysis gives real-time extracellular neurotransmitter concentration in a defined region. Post-mortem tissue analysis gives metabolite concentration. Receptor autoradiography maps distribution and density change. These are the measurements that connect a receptor event to a structural one, and a study that reports the structural change without the intermediate steps is describing a correlation.

Tianeptine purity testing in progress

What should a Tianeptine Certificate of Analysis Show?

A usable certificate names the lot, gives the test date, and reports the analytical method and its result, not just a pass mark. Nordic Chems certificates are issued per lot as PDFs containing the actual analytical graphs, so the chromatogram can be read rather than taken on trust.

Testing runs through 2 independent laboratories: BioRegen Labs in Houston, Texas, and Janoshik Analytical in Prague, Czech Republic. The methods are HPLC, mass spectrometry, and LC-MS chromatography reported with retention time and peak analysis, with heavy metals testing available on the same sample when a protocol calls for it. The purity specification is 99%. A sample that falls below it is not used, and a different lot is tested and taken forward in its place.

Matching a certificate to the material in hand is the step most researchers skip. A certificate for a different lot documents nothing about the bottle on the bench. Nordic Chems matches an order number to its lot on request by email, and the analytical documentation is published rather than issued only when asked. Incoming raw material is tested; the finished chemical is inspected physically and visually rather than retested, which is worth knowing when writing up a methods section.

Manufacturing runs under ISO 9001:2015, certificate number C2024-01140, with a scope covering custom synthesis, quality testing and distribution of high-purity chemicals. As of September 2026 the company has shipped 10,600 orders across more than a year of operation, tested 20 lots, and holds a 25% all-time repeat rate.

Handling, Storage and Containment in 2026

Store tianeptine cool, dry, and tightly closed, in an area locked or restricted to qualified personnel. Material ships induction-sealed and desiccated, and the seal is the moisture control. Refrigeration is not part of the specification, and advice to hold the material at 2 to 8 degrees Celsius in amber glass does not match how it is supplied or stabilized.

Handle with appropriate PPE and standard laboratory safety practice: gloves, eye protection, a lab coat, and weighing in a ventilated enclosure for powders. Follow applicable regulations for handling, storage, transport and disposal, and route waste through institutional chemical waste procedures with the container labeled by contents and lot. Record every weighing against the lot number in the notebook, because a certificate without a matching notebook entry cannot support a published result. Common documentation questions are answered on the supplier FAQ page.

A PERSON HOLDING RESEARCH CYLINDER

Where the US regulatory position stood in 2026

Tianeptine is not a federally scheduled controlled substance in the United States as of September 21, 2026, and it is not approved for any human use. On July 8, 2026, the DEA published a notice of proposed rulemaking to place tianeptine in Schedule I. The comment period closed August 7, 2026, with 188 comments received. No final rule had been issued at the time of writing.

This is live, and it changes what a laboratory can hold. A Schedule I placement would require a DEA research registration before acquisition or possession, which takes months. Laboratories planning work into 2027 should check docket DEA-2026-1091 before writing tianeptine into a protocol, and should not assume current availability continues.

State restrictions already exist and vary. Nordic Chems ships within the United States and Canada only, maintains shipping profiles that block restricted states at checkout, and vets each order before fulfillment, with buyers declaring they are trained professionals. Responsibility for lawful possession sits with the buyer. Confirm the position in your own state before ordering from Nordic Chems or anyone else. Any laboratory writing tianeptine into depression research through 2027 should verify both the federal docket and its own state rules first, then check the tianeptine research collection for current availability by salt form.

Disclaimer. All material described here is supplied for laboratory and educational research by qualified professionals. It is not for human or animal consumption, and it is not approved by the FDA for any human application.

FAQs

What makes tianeptine valuable for depression research compared to other compounds?

Tianeptine offers researchers a unique pharmacological profile by modulating glutamatergic neurotransmission through AMPA and NMDA receptors, unlike traditional SSRIs that target serotonin systems. Its effects on neuroplasticity, BDNF expression, and synaptic protein synthesis make it particularly valuable for investigating stress-induced neurobiological changes and alternative depression mechanisms in laboratory models.

What qualifications are required to purchase research-grade tianeptine?

Research-grade tianeptine can only be purchased by qualified researchers affiliated with accredited academic institutions, pharmaceutical laboratories, or licensed research facilities with proper documentation. Reputable suppliers like Nordic Chems require proof of professional credentials, institutional affiliation, and verification of legitimate research purposes before approving purchases.

How should research facilities store tianeptine to maintain compound integrity?

Tianeptine should be stored in temperature-controlled environments (typically 2-8°C) within sealed, amber glass containers protected from light, moisture, and oxidation. Research facilities must implement proper labeling, inventory tracking, and regular stability testing to ensure compound integrity and maintain reproducible experimental results.

What safety protocols must laboratories implement when working with tianeptine?

Laboratories must require personal protective equipment (PPE), work in well-ventilated areas or fume hoods, and maintain secure storage with restricted access to authorized personnel only. All handling must be documented in laboratory notebooks, with proper waste disposal following institutional chemical safety guidelines and OSHA standards.

Can tianeptine research findings be published in peer-reviewed journals?

Yes, tianeptine research can be published in peer-reviewed journals when studies meet rigorous methodological standards, including transparent documentation of compound source, purity verification, experimental protocols, and ethical approval for animal studies. Publications must clearly identify tianeptine as research-grade material used exclusively for laboratory investigation, contributing to scientific literature on depression neurobiology and neuroplasticity mechanisms.
